Reliable Microbial Detection with Galileo Viral Panel

Galileo™ Analytics’ foundation is built on a graph-based sequence aligner. Galileo Analytics is a cloud-based NGS bioinformatics software that fully automates the detection of microbial gene sequences, complete with integrated run- and sample-level controls. This turnkey solution provides both a report of observed microbial nucleic acid signal and a QC report per sample. Its infrastructure follows best-practice security processes and provides scalable architecture to process sequencing files quickly, reduce internal storage and organization burden.

Capturing True Signal, Not Noise

Galileo Analytics reports a signal score that correlates with quantification. This signal score weights reads by biological information content, database sequence information, alignability, and uniqueness. The signal score provides an accurate quantification unit that, when calibrated, can be converted into copies or International Units allowing validation against other quantitative standard methods.
Linearity was assessed using intact viral DNA reference materials spiked into a negative plasma matrix at 0, 1000, 5000, 10,000, and 100,000 copies or IU/ml in quadruplicate, within the same sequencing run. Linearity was demonstrated on a log-log scale. Data generated internally with paired-end sequencing on the Illumina NextSeq® Platform.

Viral Calibration Curves

Linearity was assessed using intact viral DNA reference materials spiked into a negative plasma matrix at 0, 1000, 5000, 10,000, and 100,000 copies or IU/ml in quadruplicate, within the same sequencing run. Linearity was demonstrated on a log-log scale. Data generated internally with paired-end sequencing on the Illumina NextSeq® Platform.

LOD/LLOQ Data

The lower limits of quantification (LLOQ) were determined via a Probit experiment using intact viral DNA reference materials spiked into a negative plasma matrix at 0, 1, 20, 40, 75, 150, 300, 1200, and 10,000 IU or copies/ml. The LLOQ corresponds to recovering signal with a CV value < 25%. Data generated internally with paired-end sequencing on the Illumina NextSeq® Platform.

Galileo Analytics’ integrated controls are built into the entire workflow from sample extraction to report and ensure both seamless functionality of individual components as well as facilitating the identification of errors. In addition general quality control metrics per library ensure correct processing and sequencing with easy to understand metrics. Taken together these controls aid in identifying errors and allow preventative actions to be taken.

User Friendly Reporting

The actionable reports generated by Galileo Analytics provide easy to understand ID and QC information. Quality metrics are displayed for both individual samples libraries and run-level data. The QC Reports accurately detail the performance of the assay controls and provide ranges for reference allowing easy monitoring of each step of the workflow.
